The regulation of tolerance to self-antigens is a necessary biological process in order to prevent autoimmunity. CD4+ CD25+ regulatory T cells are known regulators of immune tolerance and activation. In both human and mice species, CD4+ CD25+ regulatory T cells are consistent in their expression of the transcription factor FOXP3 (forkhead box P3). FOXP3 acts as the “master switch” for the function and development of regulatory T cells. Mutations and deletions in the sequence encoding for FOXP3 expression lead to an inability to express FOXP3, causing an inability to regulate T cell development and leading to fatal autoimmune diseases such as IPEX and XLAAD. Conversely, recent studies have shown high and wide expression of FOXP3 in several cancer cell lines and tumors, indicating that FOXP3 is involved in preventing immunity against malignant tumors.

This antibody recognizes the N-terminus of Forkhead box protein P3 (FOXP3).
The protein encoded by this gene is a member of the forkhead/winged-helix family of transcriptional regulators. Defects in this gene are the cause of immunodeficiency polyendocrinopathy, enteropathy, X-linked syndrome (IPEX), also known as X-linked autoimmunity-immunodeficiency syndrome. Alternatively spliced transcript variants encoding different isoforms have been identified.
FUNCTION: Probable transcription factor. Plays a critical role in the control of immune response.
SUBCELLULAR LOCATION: Nucleus Potential.
TISSUE SPECIFICITY: High level of expression in thymus and spleen.
INVOLVEMENT IN DISEASE: Defects in Foxp3 are the cause of the scurfy phenotype (sf). It results in a lethal disorder of immunoregulation, characterized by infections, diarrhea, anemia, thrombocytopenia, hypogonadism, gastrointestinal bleeding, lymphadenopathy and leukocytosis.
Expected molecular weight ~47 kDa. On western blot, a molecular weight of ~49-55 kDa may be observed (Bolzer, K., et al., 2009).
