Background InformationProtein Gene Product 9.5 (PGP9.5) is an abundant protein in many tissues, but especially so in neurons where it has been effectively used as a phenotypic marker and is immunologically distinct from neuron specific enolase. The 27 kDa protein was first identified by high resolution two dimensional PAGE. Standard immunohistochemical techniques have demonstrated the presence of PGP9.5 in neurons and nerve fibers at all levels of the central and peripheral nervous system, in many neuroendocrine cells, in segments of the renal tubules, in spermatogonia and Leydig cells of the testis, in ova and in some cells of both the pregnant and non-pregnant corpus luteum. PGP9.5 is known to be a member of the ubiquitin C-terminal hydrolase family and immunohistochemical studies have shown that the protein is enriched in several ubiquinated inclusion bodies, suggesting that such structures may be metabolically dynamic regions of the cell.

Entrez Gene Number
Entrez Gene SummaryUCHL1 is a member of a gene family whose products hydrolyze small C-terminal adducts of ubiquitin to generate the ubiquitin monomer. Expression of UCHL1 is highly specific to neurons and to cells of the diffuse neuroendocrine system and their tumors. It is present in all neurons (Doran et al., 1983 [PubMed 6343558]).[supplied by OMIM]

UniProt Number
UniProt SummaryFUNCTION: SwissProt: P09936 # Ubiquitin-protein hydrolase involved both in the processing of ubiquitin precursors and of ubiquitinated proteins. This enzyme is a thiol protease that recognizes and hydrolyzes a peptide bond at the C-terminal glycine of ubiquitin. Also binds to free monoubiquitin and may prevent its degradation in lysosomes. The homodimer may have ATP-independent ubiquitin ligase activity.
SIZE: 223 amino acids; 24824 Da
SUBUNIT: Homodimer. Interacts with SNCA (By similarity). Interacts with COPS5.
SUBCELLULAR LOCATION: Cytoplasm.
TISSUE SPECIFICITY: Found in neuronal cell bodies and processes throughout the neocortex (at protein level). Expressed in neurons and cells of the diffuse neuroendocrine system and their tumors. Weakly expressed in ovary.
PTM: O-glycosylated (By similarity).
DISEASE: SwissProt: P09936 # Oxidation of Met-1, Met-6, Met-12, Met-124 and Met-179 to methionine sulfoxide, and oxidation of Cys-220 to cysteine sulfonic acid have been observed in brains from Alzheimer disease (AD) and Parkinson disease (PD) patients. In AD, UCHL1 was found to be associated with neurofibrillary tangles.
SIMILARITY: SwissProt: P09936 ## Belongs to the peptidase C12 family.
MISCELLANEOUS: In contrast to UCHL3, does not hydrolyze a peptide bond at the C-terminal glycine of NEDD8.
Molecular Weight24-27 kDa

Usage Statement
  • Unless otherwise stated in our catalog or other company documentation accompanying the product(s), our products are intended for research use only and are not to be used for any other purpose, which includes but is not limited to, unauthorized commercial uses, in vitro diagnostic uses, ex vivo or in vivo therapeutic uses or any type of consumption or application to humans or animals.
Storage and Shipping Information
Storage ConditionsStable for 1 year at -20°C in undiluted aliquots (not less than 10 μL) from date of receipt. The use of high quality “antiserum-grade” plastic or glass vials is recommended.

Handling Recommendations:
Upon first thaw, and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
