Background InformationNO, produced by nitric oxide synthases (NOS), is a critical signaling molecule involved in many physiological processes including muscle contraction, vasodilation, neuronal transmission, and immune responses. Some of these effects are mediated through reaction with and activation of guanylate cyclase (GC), resulting in increased cGMP production. With a half-life of seconds, NO readily diffuses through the cytosol and cell membrane, allowing it to act inside or adjacent to the cell in which it was produced.

Biological Information
ImmunogenKLH-conjugated, synthetic peptide (C-RIRTQ[pS]FSLQER) corresponding to amino acids 1172-1183 of human eNOS (endothelial nitric oxide synthase) with an N-terminal cysteine added for conjugation purposes. The immunizing sequence is identical in rat, mouse, bovine, pig, and dog.
EpitopeSer1177
ConcentrationPlease refer to the Certificate of Analysis for the lot-specific concentration.
HostRabbit
SpecificityRecognizes phosphorylated eNOS, Mr 132 kDa. Additional unknown bands at 50 and 64 kDa were competed by the immunizing peptide.

UniProt Number
UniProt SummaryFUNCTION: SwissProt: P29474 # Produces nitric oxide (NO) which is implicated in vascular smooth muscle relaxation through a cGMP-mediated signal transduction pathway. No mediates vascular endothelial growth factor (VEGF)-induced angiogenesis in coronary vessels and promotes blood clotting through the activation of platelets.
COFACTOR: Heme group. & Binds 1 FAD. & Binds 1 FMN. & Metrahydrobiopterin (BH4). May stabilize the dimeric form of the enzyme.
SIZE:1203 amino acids; 133289 Da
SUBUNIT: Homodimer. Interacts with NOSIP and NOSTRIN.
SUBCELLULAR LOCATION:Cell membrane, caveola. Cytoplasm, cytoskeleton. Golgi apparatus. Note=Specifically associates with actin cytoskeleton in the G2 phase of the cell cycle; which is favored by interaction with NOSIP and results in a reduced enzymatic activity.
TISSUE SPECIFICITY: Platelets, placenta, liver and kidney.
SIMILARITY: SwissProt: P29474 ## Belongs to the NOS family. & Contains 1 flavodoxin-like domain.
Molecular Weight132 kDa

Usage Statement
  • Unless otherwise stated in our catalog or other company documentation accompanying the product(s), our products are intended for research use only and are not to be used for any other purpose, which includes but is not limited to, unauthorized commercial uses, in vitro diagnostic uses, ex vivo or in vivo therapeutic uses or any type of consumption or application to humans or animals.
Storage and Shipping Information
Storage ConditionsStable for 1 year at -20°C from date of receipt.
For maximum recovery of product, centrifuge the vial prior to removing the cap.
Handling Recommendations: Upon first thaw, and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance. Note: Variabillity in freezer temperatures below -20°C may cause glycerol containing solutions to become frozen during storage.
