Drosophila eyes shut protein is expressed in photoreceptor cells, the central nervous system, and peripheral sensory ganglia, and is essential for the formation of a matrix-filled intra-rhabdomeral space within the fly's retina ommatidia, the loss of spacemaker (spam) converts an open rhabdom system to a closed one. Human EYS/SPAM gene likewise is found to be expressed in the eye and the spinal cord, and mutations in human EYS result in retinitis pigmentosa. Human EYS protein contains 27 EGF-like repeats and five laminin G-like (LG1-5) domains (between EGF repeats 21 & 21, 21 & 22, 23 & 24, 25 & 26, and after repeat EGF repeat 27). The presence of the EGF-like repeats is indicative of a role in the Notch signaling pathway, while EYS LG1-5 bear resemblance to the three agrin laminin G domains that are known to interact with several proteins, including α-dystroglycan at the muscle surface.
