GPAT4 is a member of the 1-acylglycerol-3-phosphate O-acyltransferase family that plays a role in triglyceride biosynthesis in several tissues. It is a rate-limiting enzyme that converts glycerol-3-phosphate (G3P) to 1-acyl-sn-glycerol-3-phosphate by incorporating an acyl moiety at the sn-1 position of the glycerol backbone. It is shown to be active against both saturated and unsaturated long-chain fatty acyl-CoAs. Its HXXXXD motif (aa 248-253) is reported to be essential for its acyltransferase activity and may constitute the binding site for the phosphate moiety of the glycerol-3-phosphate. By increasing triglyceride synthesis, it protects cells against lipotoxicity induced by high levels of saturated fatty acids. (Ref.: Piccolis, M., et al. (2019). Mol.
