Background InformationHuman Rad18 (hRad18) is a homologue of the S. pombe Rad18. In yeast, Rad6 and Rad18 play a crucial role in post−replication repair functions in gap−filling of daughter strands on replication of damaged DNA. Furthermore, Rad6−Rad18 interaction is thought to be critical at an early step during lesion bypass in yeast. In vivo, Rad18 protein forms a tight complex with Rad6 through a conserved ring−finger motif. It has been suggested that the Rad18−Rad6 complex binds to the damaged sites by the single−stranded DNA binding property of Rad 18. This is followed by degradation of the stalled components of DNA replication by the ubiquitin−conjugating enzyme activity of Rad6. The human Rad18 maps on chromosome 3p24−25, the region which is often deleted in lung, breast, ovary, and testis cancers. The human RAD18 gene codes for a protein of 495 amino acids with a predicted molecular weight of 56 kDa. The human Rad18 is ubiquitously expressed in various human tissues.

Application NotesWestern Blotting Analysis: A representative lot detected an upregulated RAD18 in the chromatin fraction from mouse embryonic fibroblasts (MEFs) following DNA damage induction by UV irradiation or mitomycin C (Cat. Nos. 475820 & 47589) treatment. DNA damage induced RAD18 chromatin association was not observed in Brca1-/- MEFs (Tian, F., et al. (2013). Proc. Natl. Acad. Sci. U. S. A. 110(33):13558-13563).
Western Blotting Analysis: Representative lots detected RAD18 expression in wild-type mouse embryonic fibroblasts (MEFs), while reduced expression was seen in Rad18+/- MEFs, and no expression was detected in Rad18-/- MEFs (Sun, J., et al. (2009). Mech. Dev. 126(3-4):173-183; Tateishi, S., et al. (2003). Mol. Cell. Biol. 23(2):474-481).
Immunocytochemistry Analysis: A representative lot detected a large reduction of UV-induced RAD18 nuclear foci formation in Brca1-/- mouse embryonic fibroblasts (MEFs) when compared with wild-type MEFs by fluorescent immunocytochemistry staining of methanol-fixed cells (Tian, F., et al. (2013). Proc. Natl. Acad. Sci. U. S. A. 110(33):13558-13563).
Immunocytochemistry Analysis: A representative lot detected RAD18 immunoreactivity in 3.7% formaldehyde- and 80% methanol-fixed wild-type mouse embryonic fibroblasts (MEFs) by fluorescent immunocytochemistry, while only background staining was seen in Rad18-/- MEFs (Tateishi, S., et al. (2003). Mol. Cell. Biol. 23(2):474-481).
Immunohistochemistry Analysis: A representative lot detected nuclear Rad18 immunoreactivity among undifferentiated spermatogonia (germinal stem cells) in 4% paraformaldehyde-fixed, methyl methacrylate-embedded testes sections from one- and six-week old mice (Sun, J., et al.
