Immunocytochemistry Analysis: A 1:250 dilution of this antibody detected Histone H3 (Met3-Lys9) in Mouse Embryo Fibroblasts (MEF).
Peptide Inhibition Assay: Target band detection in acid extract of HeLa cells was prevented by pre-blocking of a representative lot with the immunogen Histone H3 (Met3-Lys9) peptide, but not the corresponding unmodified Histone H3 peptide.
Dot Blot Analysis: A 1:1,000 dilution from a representative lot detected trimethyl-Histone H3 (Lys9) peptide, but did not detect di-methyl Histone H3 (Lys9) peptide, mono-methyl Histone H3 (Lys9) peptide, nor unmodified Histone H3 (1-19) peptide.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

General description
Histone H3 is one of the five main histone proteins involved in the structure of chromatin in eukaryotic cells. Featuring a main globular domain and a long N-terminal tail, H3 is involved with the structure of the nucleosomes of the ′beads on a string′ structure. The N-terminal tail of histone H3 protrudes from the globular nucleosome core and can undergo several different types of epigenetic modifications that influence cellular processes. These modifications include the covalent attachment of methyl or acetyl groups to lysine and arginine amino acids and the phosphorylation of serine or threonine.

Preparation Note
Store at -10°C to -25°C. Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
