アムホテリシン B は、ストレプトマイセス属由来のポリエン抗真菌抗生剤です。ステロール、特に、真菌と細菌両方の細胞膜のエルゴステロールに高い親和性を持ちます。 細胞培養での抗真菌活性の維持において、通常は 2.5 mg/L で培地に使用され、ペニシリンとストレプトマイシンと併用されます。

Biochem/physiol Actions

アムホテリシン B はステロールと結合して、膜内に細孔を形成し、低分子の漏れ出しを引き起こします。本製品は、真菌や酵母に対して最も効果的です。

Disclaimer

アムホテリシン B は、37°Cの培養で 3 日間、活性を維持します。空気と光から保護し、-20°Cで保存した場合、本製品は 5 年間活性を維持します。

容器を密閉し、乾燥した風通しの良い場所で保管してください。製品は光と湿気の影響を受けます。乾燥した場所に保管します。
水で溶解すると、本製品は溶解補助剤のデオキシコール酸を含むアムホテリシンBの懸濁液になります。 凍結乾燥製剤は、リン酸ナトリウムと塩化ナトリウム中に約45％のアムホテリシンBと35％のデオキシコール酸ナトリウムが含まれています。

可溶化アムホテリシンBはアンバーのガラス瓶に充填されています。

Preparation Note

アムホテリシン B は、中性 pH において 19.6～20.4 mg/mL の濃度で加熱せずに水に可溶であり、黄色く透明な、またはわずかに濁った溶液になります。
溶解度が低いため、水溶液は濾過滅菌できません。 滅菌水に再溶解する場合、濾過滅菌する必要はありません。

また、DMSO（30～40 mg/mL）やジメチルホルムアミド（2～4 mg/mL）にも溶解します。
