General description
Phospho-α-synuclein (S129) is a modified form of the protein α-synuclein, characterized by the phosphorylation at serine 129. This post-translational modification influences the protein′s structure and function, promoting its aggregation into toxic fibrils, which are a hallmark of several neurological disorders. In healthy neurons, α-synuclein plays a role in synaptic function, regulating neurotransmitter release and maintaining synaptic integrity. However, when phosphorylated at S129, α-synuclein tends to misfold and aggregate, leading to the formation of Lewy bodies, which are implicated in neurodegenerative diseases such as Parkinson′s disease and dementia with Lewy bodies.
Phospho-α-synuclein (S129) also contributes to neuroinflammation by activating microglial cells, which can exacerbate neuronal damage and promote disease progression. Its presence in the brain serves as a biomarker for neurodegeneration, highlighting its critical role in the pathophysiology of various neurodegenerative conditions.
