Gene Information
human ... RAD21(5885)
General description
100-120kDa
Aka Scc1, part of the cohesin complex, Rad21 is a nuclear protein that associates with mitotic chromatin and is essential for the association of kinetochores with microtubules. Rad21/Scc1 s is cleaved by caspases, promoting apoptosis.
The protein encoded by the Rad21 gene is highly similar to the gene product of Schizosaccharomyces pombe rad21, a gene involved in the repair of DNA double-strand breaks, as well as in chromatid cohesion during mitosis. This protein is a nuclear phospho-protein, which becomes hyperphosphorylated in cell cycle M phase.
RAD21 also plays a role in apoptosis, via its cleavage by caspase 3 or caspase 7 during early steps of apoptosis: the C-terminal 64 kDa cleavage product may act as a nuclear signal to initiate cytoplasmic events involved in the apoptotic pathway. Recent work has shown that the RAD21 has a role in the invasion and metastasis of cancer cells

The spatial organization, correct expression, repair, and segregation of eukaryotic genomes depend on cohesin, ring-shaped protein complexes that are thought to function by entrapping DNA It has been proposed that cohesin is recruited to specific genomic locations from distal loading

The end joining of distant DNA double-strand ends (DSEs) can produce potentially deleterious rearrangements. We show that depletion of cohesion complex proteins specifically stimulates the end joining (both C-NHEJ and A-EJ) of distant, but not close, I-SceI-induced DSEs in S/G2
