General description
220 kDa
BRCA1とBRCA2という2つの主要乳がん感受性遺伝子が確認されています。両遺伝子とも腫瘍抑制遺伝子と考えられています。BRCA1およびBRCA2内の変異は、ほとんどの家族性乳がん症例の原因となっています。BRCA1およびBRCA2遺伝子産物の機能解析により、転写制御とDNA損傷修復への二重の関与が立証されています。 BRCA1は細胞周期の制御に影響を及ぼし、その発現低下によるBRCA1の機能喪失は、p53およびp21遺伝子を介して細胞周期の停止につながります。In vivoでは、BRCA1とBRCA2は急速に増殖する細胞で最大レベルで発現しています。この特徴は、BRCA1とBRCA2が細胞周期依存的に発現するというin vitroでの観察と一致します。乳腺発生中、BRCA1とBRCA2の発現は急速に増殖する細胞区画で誘導されます。
