Biochem/physiol Actions

β/A4アミロイド前駆体タンパク質（APP）は、タンパク質分解切断を受け、β/A4ペプチドを生成します。このペプチドの細胞外沈着が、脳皮質実質および脳髄膜血管で認められており、これがアルツハイマー病の病的状態の特徴となっています。APP遺伝子の変異は、アルツハイマー病の早期発症やアミロイドーシスを伴う遺伝性脳出血を引き起こします。
抗APP A4抗体は、pre-A4と反応し、pre-A4分子のN末端のアミノ酸66-81を認識します（Hilbich et al., 1993)。 22C11は、APPの3つのアイソフォーム、すなわち未成熟の約110 kDa、sAPP 約120 kDa、および成熟した約130 kDaをすべて認識します(Hoffmann et al., 2000)。抗APP A4抗体は、APLP2と交差反応することが知られています（Slunt, 1994)。

General description

β/A4 アミロイド前駆体タンパク質（APP）は膜貫通糖タンパク質であり、3つのアイソフォームとして存在します。このタンパク質は、1回膜貫通ドメイン、大型の細胞外N末端ドメイン、および短い細胞質C末端尾部で構成されています。APP遺伝子はヒト染色体の21q21.3-q22上に存在します。
110/120/130 kDa。
