General description
2,3,5-トリフェニルテトラゾリウムクロリド(TTC)は、主に固体培地中の微生物コロニーの簡単な菌数測定に使われる色素です。この色素は酸化型では無色ですが、微生物による還元でホルマザンが生成されると赤色に変わります。E. coliなどの大腸菌群のような生きた微生物が酵素作用でTTCを還元することで赤色のホルマザンが生成され、細胞の顆粒内に残ります。低温殺菌牛乳ではTTCを還元できない微生物の割合が高くなりますが、検査室の手順ではそのことに気づけないため、TTCを用いた低温殺菌牛乳の検査を強くお勧めします。高濃度のTTCには、グラム陽性菌の増殖を阻害する効果がある可能性があります。
Application
2,3,5-トリフェニルテトラゾリウムクロリド(TTC)溶液は、細胞呼吸があることを示して生細胞と死細胞を見分けたり、固体培地でTTCを還元できる微生物を同定したりするのに役立つ、酸化還元指示薬です。
