General description

グラム染色キットは、すべての微生物検査の専門ラボにおいて最も広く採用されている鑑別染色技術で用いられています。あらゆる種類の細菌分離株に用いる同定法において最も重要な基準の1つです。グラム陽性菌とグラム陰性菌の細胞壁には多くの生理学的相違点があります。クリスチャン・グラムによるグラム染色の発見以降、このプロセスは広く検討され、再定義されてきました。実用上は、細菌細胞の薄い塗抹標本をクリスタルバイオレットで染色した後、一次染色の結合を強化するために媒染剤を含むヨウ素で処理します。アルコールまたはアセトンの脱色液を用いて、弱く結合している細胞からクリスタルバイオレットを除去した後、サフラニンの対比染色液を用いて、脱色した細胞に色のコントラストをつけます。グラム染色法によって適切に染色された塗抹標本では、グラム陽性菌は青色～紫色に見え、グラム陰性菌はピンク色～赤色に見えます。

Application

グラム染色法は、微生物検査における最も重要な染色技術です。グラム陽性菌とグラム陰性菌を区別するためのあらゆる種類の細菌分離株の同定法において、最も重要な基準でもあります。
