Application
IPTGは、β-ガラクトシダーゼ活性の発現誘導が必要なクローニングの際に用いられます。大腸菌中のlacオペロンの発現を誘導させる組み換えコロニーのblue-white selectionの際に、X-GalまたはBluo-Gal とともに用います。β-ガラクトシダーゼをコードしているlacZ遺伝子を抑制しているlacIリプレッサーに結合してコンホメーションを変化させ、その抑制を妨げます。
Preparation Note
IPTGを水に溶解しフィルター滅菌してストック溶液 (0.1 M)を作成します。プレート中の最終濃度は0.2 mMです。
