Application

ウサギで産生された抗アクチン、N末端抗体は、
  • イムノブロッティング
  • 定量的クロマチン免疫沈降（qChIP）に使用されています。

ウサギ抗アクチン、N末端抗体は、ウェスタンブロッティングアッセイに使用できます。本抗体は、培養ニワトリ線維芽細胞を使用した1～2 μg/mLでの免疫細胞染色にも使用できます。さらに、本抗体は、ヒトの虫垂、マウスの心臓、およびカエルの骨格筋の切片を使用して、2～4 μg/mLの濃度で免疫組織染色で使用するのに適しています。
胚性マウス肢組織から生成したたんぱく質溶解物が、アクチンを対照物質としてウェスタンブロットにより解析されました。アクチンは、ウサギ抗アクチンを用いて1:8000で検出されました。ウサギ抗アクチン抗体によって検出されたアクチンが、MCF7細胞溶解産物の対照物質として使用されました。アクチンは42 kDaで検出されました。

Biochem/physiol Actions

アクチンとミオシンは多くのタイプの細胞の構成要素であり、細胞形状の維持、運動、分泌、細胞質流動、食作用、および細胞質分裂など種々の細胞プロセスに関与しています。アクチンのN末端ドメインは、分子の主要な抗原領域であるようにみえます。異なるアイソフォームは同じ細胞内で共存できますが、異なって制御されており、ほとんどの場合、機能的に互いと置き換わることはできません。

General description

アクチンは、最も保存されている真核生物タンパク質の1つであり、哺乳動物および鳥類で少なくとも6通りの主要な異なるアイソフォームとして発現します。うち4つのアイソフォームは、筋組織を区別するマーカーであり、そのうちの2つはほぼすべての非筋細胞内に認められます。アクチンのアイソフォームは、全体では90%以上の配列相同性を示しますが、N末端の18残基では50～60%の相同性を示します。アクチンは、球状単量体（Gアクチン）として、また線維状アクチン（Fアクチン）の重合体としてこれらの両方が細胞内に存在しており、さまざまな安定および不安定な構造体の形成に関与しています。細胞核内および細胞膜内にアクチンが存在することが報告されています。
アクチンは、細胞の運動性、分泌、食作用および細胞質分裂を調節する細胞骨格タンパク質です。アクチンのNH2-末端は、抗原として機能する場合があります。この末端は、アクチン相互作用を調節し、ミオシンのようなタンパク質と会合することもできます。

N末端側アクチン9アミノ酸に相当する合成ノナペプチドのKLH結合体。
