アムホテリシンBは、親水性ポリヒドロキシル鎖1本と親油性ポリエンをベースとする炭化水素鎖1本を有する大環状ラクトンです。

Application

アムホテリシンB溶液は以下の用途に使用されています：

マウスのAspergillus fumigatus（アスペルギルス・フミガーツス）感染の治療ための腹腔内注射剤として
中国のメリノの雌羊の卵巣を培養するための吸引培地の成分として
ウマの黄体採取中の輸送培地の成分として
マウス－マウスB細胞ハイブリドーマ55-6細胞株を培養するためのPRMI培地のサプリメントとして

Biochem/physiol Actions

アムホテリシンBは、全身的な真菌感染の治療に臨床的に使用される、Streptomyces（ストレプトマイセス）sp.由来のポリエン系の抗真菌抗生剤です。真菌および細菌の細胞膜のステロール、特にエルゴステロールに対して高い親和性を有します。抗真菌での細胞培養の維持に通常使用される濃度は2.5 mg/Lであり、ペニシリンおよびストレプトマイシンと共に培地に添加して使用します。

アムホテリシンBは、真菌や酵母に対して最も効果的です。本製品は、膜を伴うステロールと結合することによって膜貫通路を形成し、その結果として、低分子物質の漏出を誘導します。アムホテリシンBは、真菌症の治療に対する有効な抗生剤ですが、毒性も発現します。

Preparation Note

通常、本物質が水溶液中に沈殿を形成することはありません。溶液を混合することで大抵の沈殿は再溶解できますが、完全に溶解できない場合でも、アムホテリシンBはまだ使用できます。
