Application

アプロチニンは、トリプシンの阻害剤として主に使用されます。
ウシ肺由来のアプロチニンは、インスリンおよびグルカゴン様ペプチド-1アッセイに使用されています。ウエスタンブロッティングにも使用されています。

Biochem/physiol Actions

アプロチニンは58アミノ酸の天然ポリペプチドです。止血／抗プラスミン作用があり、手術中の出血を抑えるのに役立ちます。そのため、輸血の必要がなくなります。
アプロチニンは競合的セリンプロテアーゼ阻害剤で、酵素と安定な複合体を形成し活性部位をブロックします。この結合は可逆的で、ほとんどのアプロチニン‐プロテアーゼ複合体は pH が 10 を超えるか、または 3 を下回るような極端な pH レベルでは解離します。アプロチニンは、構造的には、58 アミノ酸からなるウシ肺由来のグロブリン様タンパク質モノマーであり、単一のポリペプチド鎖中の 3 つのジスルフィド結合によって架橋されています。

Other Notes

1 トリプシン阻害剤ユニット（TIU）は、2 トリプシン単位の活性を 50％減少させ、1 トリプシン単位は、pH 7.8 および 25°Cにおいて、1 分間で 1 μ モルの N-α-ベンゾイル-DL-アルギニンp-ニトロアニリドを加水分解します。他にも一般的に使用される単位として、KIU があります。1 TIU = 1,300 KIU で換算します。
ベンゾイル-L-アルギニンエチルエステル（BAEE）を担体として使用する場合、BAPNAの1ユニットは約25°C、pH8.0で約45BAEE μモルユニット、または25°C、pH7.6で約9,000 BAEE A253ユニットです。
