ブタ膵臓から分離されるα-アミラーゼは糖タンパク質です。残基数475個未満の単一ポリペプチド鎖であり、SH基2個およびジスルフィド架橋4個を有し、Ca2+が強く結合して安定化に寄与しています。塩素イオンは活性と安定に必要とされています。活性のpH範囲は5.5～8.0であり、至適pHは7です。

Application

α-アミラーゼは、デンプンやグリコーゲンなどのα-結合を持った多糖類中のα結合の加水分解に使用されます。Sigmaのα-アミラーゼは、シロイヌナズナの代謝研究などの、様々な植物研究で使用されています。

Biochem/physiol Actions

α-アミラーゼは、3つ以上のα-(1,4)-D-グルコースの多糖中のα-(1,4)グルカン結合を加水分解します。デンプンやグリコーゲンなどの天然基質は、グルコースとマルトースに分解されます。ブタ膵臓由来のα-アミラーゼは、2つのSH基と4つのジスルフィド結合を含む約475残基のポリペプチド鎖1本と、安定性のために必要な密に結合したCa2+からなる糖タンパク質です。
