アプロチニンは競合的セリンプロテアーゼ阻害剤で、酵素と安定な複合体を形成し活性部位をブロックします。この結合は可逆的で、ほとんどのアプロチニン‐プロテアーゼ複合体は pH が 10 を超えるか、または 3 を下回るような極端な pH レベルでは解離します。アプロチニンは、構造的には、58 アミノ酸からなるウシ肺由来のグロブリン様タンパク質モノマーであり、単一のポリペプチド鎖中の 3 つのジスルフィド結合によって架橋されています。

1 トリプシン阻害剤ユニット（TIU）は、2 トリプシン単位の活性を 50％減少させ、1 トリプシン単位は、pH 7.8 および 25°Cにおいて、1 分間で 1 μ モルの N-α-ベンゾイル-DL-アルギニンp-ニトロアニリドを加水分解します。他にも一般的に使用される単位として、KIU があります。1 TIU = 1,300 KIU で換算します。
1トリプシンインヒビターユニット(TIU)は、2トリプシンユニットの活性を50%低減させる量です(1トリプシンユニットはpH 7.8、25°C、1分間で1.0 μmolのN-α-ベンゾイル-DL-アルギニンp-ニトロアニリド(BAPNA)を加水分解する酵素量)。
