General description

アキュターゼ®溶液［Ca++またはMg++を含まないダルベッコリン酸緩衝生理食塩水（DPBS）］は、細胞剥離のためのタンパク質分解酵素およびコラーゲン分解酵素を含有しています。標準的な組織培養プラスチック製品や接着コーティングされたプラスチック製品からの細胞剥離に有用です。アキュターゼは、細胞を少ない損傷で培養容器から剥離するために役立つため、細胞剥離に関してはトリプシンよりも推奨されます。そのため、アキュターゼは、感受性細胞の処理や表面マーカーの分析のためによく用いられます。

Application

アキュターゼ®溶液は、脳腫瘍細胞、人工多能性幹細胞（IPSC）および膠芽腫細胞を単一細胞に分散させるなど、凝集したクラスターの中で増殖する非付着細胞の剥離や消化に使用されています。

シーケンシングのための単一細胞懸濁液の調製用に適しています。
一次線維芽細胞、内皮細胞、ニュ-ロン、腫瘍細胞株、昆虫細胞の剥離に有効であることが証明されています。 細胞表面マ-カ-の分析、ウイルス成長アッセイ、フロ-サイトメトリ-およびバイオリアクタ-のスケ-ルアップに使用する細胞の剥離に、非常に有用です。 哺乳類または微生物に由来する産物を含んでいません。

Features and Benefits

タンパク質分解酵素とコラ-ゲン分解酵素からなる独自組成の混合液です。標準的なプラスチック容器や接着被膜されたプラスチック容器からの細胞の剥離にそのままお使いいただけます。

Preparation Note

0.5 mM EDTA•4Na、3 mg/L フェノールレッド含有ダルベッコPBS(0.2 g/L KCl、0.2 g/L KH2PO4、8 g/L NaCl、1.15 g/L Na2HPO4)で調製。製品の使用法情報ウェブサイト上の製品データシートをご参照ください。
本製品は、フェノールレッドを含んでいます。 本製品は、ドライアイスと共に出荷されるため、パッケージ内に相当量の CO2 が蓄積することがあります。この CO2 は、溶液内に侵入し、わずかに pH を下げ、黄色-オレンジ色（pH 6.8）を生成することがあります。この溶液はそのままでも使用できますが、pH を調整するために水酸化ナトリウムを添加しても構いません。完全に凍結した Accutase のボトルは黄色ですが、解凍するとオレンジ色に変化します。一度開封した製品は、再凍結することも、直接冷蔵庫に入れて後で使用することもできます。 暖かいウォーターバスでこの製品を解凍ないでください。Accutase は 37 °C 以上の温度に敏感であり、37 °C で 45 分間置くと不活性化します。冷蔵庫で一晩かけて解凍するか、1～1.5 時間ほど水道水の入った容器で解凍することをお勧めします。 解凍後は、成分が均一に溶解するように、Accutase のボトルを振ってください。この製品は、使用前に温める必要はありません。冷蔵庫から取り出したら速やかに使用し、使用後は冷蔵庫に戻してください。
