General description
α-アミラーゼは主として唾液腺および膵臓で大量に産生されます。アミラーゼの発現は空腸および乳腺でも見られます。α-アミラーゼをコードする遺伝子は、1番染色体短腕に位置するAMY1およびAMY2です。AMY1は唾液および乳腺中でのアミラーゼの産生を担い、AMY2は膵臓でアミラーゼを合成させます。AMY1の発現は一定の腫瘍組織でも生じます。哺乳類のアミラーゼは、3つの構造的ドメイン（A、B、C）で構成されています。ドメインAには、2つのアルパラギン酸残基と1つのグルタミン酸残基がある活性部位が含まれます。ドメインBは活性部位領域に接し、タンパク質高次構造を維持するために必要です。ドメインCは触媒機構に関与します。

Application
ウサギで産生された抗a-アミラーゼ抗体は、免疫蛍光分析および免疫組織化学的分析に使用されています。
ウサギで産生される抗α-アミラーゼ抗体も、免疫細胞化学分析に使用されています。
マウス膵臓切片を4％パラホルムアルデヒドで固定し、1：100希釈のウサギ抗アミラーゼ抗体を用いた免疫組織化学で使用されました。
Biochem/physiol Actions
α-アミラーゼは、ヒトの食物中の主な炭水化物源であるスターチの消化の第一段階を触媒するために必要です。α-アミラーゼは多糖を加水分解し、最終的に主な分解産物としてマルトース、マルトトリオースおよび限界デキストリンを生成します。
抗血清は、ヒト唾液およびヒト膵臓抽出物に含まれるヒトα-アミラーゼに特異的です。他のヒト唾液タンパク質または膵臓抽出物タンパク質との反応は観察されません。
