Biochem/physiol Actions

プロテインアルギニンN-メチルトランスフェラーゼ(PRMT)は、タンパク質輸送、シグナル伝達や転写調節に関与する翻訳後修飾に関わっています。AMI-1は、AdoMet結合部位について競合するメチルトランスフェラーゼ阻害剤の多くとは異なり、リジンメチルトランスフェラーゼ活性を阻害せず、S-アデノシルメチオニン(AdoMet)と相互作用しません。AMI-1は、核内受容体に制御されるエストロゲンおよびアンドロゲン応答エレメントからの転写を調節することができ、HIV-1逆転写酵素阻害剤として作用します。AMI-1は、NADPH酸化酵素に由来するスーパーオキシド産生の強力なアンタゴニストですが、メチルトランスフェラーゼの阻害を介して間接的に作用するのではなく、直接的に抗酸化剤として作用します。
AMI-1 inhibits arginine, but not lysine, methyltransferase activity in vitro, while not interacting with S-adenosyl methionine (AdoMet), unlike most methyltransferase inhibitors which compete for the AdoMet binding site.
