General description

ウシ血清アルブミンは、血液の膠質浸透圧の調節を主要な生物学的機能とするタンパク質です。BSAは、さまざまな研究や用途に幅広く使用されています。BSAは、583個のアミノ酸から構成される水溶性のタンパク質で、計算された分子量は66,430ダルトンです。6つのαヘリックスが、BSAの3つの相同性ドメインを形成しています。pH依存的に可逆的な立体構造的異性化を受けます。ネイティブなタンパク質の構造は、加熱によって反応性および柔軟になります。

Preparation Note

ヒートショック分画法を用いて調製
血清アルブミンはフラクション（画分）Vと呼ばれる場合があります。この命名規則は、冷エタノール沈殿を用いて血清タンパク質を分画するオリジナルのCohn（コーン）法から取られています。血清アルブミンは、Cohn法を用いて第5エタノール画分に認められました。それ以来、「フラクションV」という用語は、調製の方法に関係なく血清アルブミンを記述するのに使用されています。他の人はこの用語を使用して、オリジナルのCohn法が記述されて以降大幅に改変されたエタノール分画法により精製された血清アルブミンのことを記述しています。Sigma-Aldrichは、真のCohn分画法、改変エタノール分画法、熱ショックやクロマトグラフィーなど様々な主要方法で精製された血清アルブミンを製造および販売しています。さらなる精製ステップには、結晶化や活性炭ろ過が含まれる場合があります。
