General description

トリメチルグリシンまたはN,N,Nトリエチルアンモニウムアセテートとも呼ばれるベタインは、3つのメチル基を持つグリシンの類似体です。ベタインのポリメラーゼ連鎖反応（PCR）バッファー混合液との適合性は高いです。ベタインはPCR産物の収率および特異性を改善するために広く用いられているPCR促進試薬であり、特にGC含量の高い標的配列や、二次構造を形成して収率が低下する標的配列のPCR増幅に有効です。ベタインは、DNA鎖の分離を促進し、DNAのGC対とAT対間のDNA融解温度（Tm）差を調整します。GC塩基対とAT塩基対の寄与を同等にすることで、dsDNAを安定化します。ベタインは、マルチプレックスおよび「長く正確な」ポリメラーゼ連鎖反応（LA-PCR）を最適化するために広く使用されています。PCR混合液に1.0～1.7 Mのベタイン溶液を添加することで、DNA鎖の融解温度の塩基対組成への依存度が低減されると報告されています。
