Biochem/physiol Actions
脳由来神経栄養因子(BDNF)は13.6 kDa(または27.2 kDa二量体)で、ニュ-ロトロフィンファミリ-に属しています。BDNFは、ヒト、マウス、ブタで同一のアミノ酸配列を有し、完全な交差反応を起こします。BDNFは、中枢神経系の神経細胞群やそこに直接結合する細胞群の発生や維持に重要です。BDNFは、神経堤、プラコ-ド由来感覚ニュ-ロン、黒質ド-パミン作動性ニュ-ロン、前脳基底部コリン作動性ニュ-ロン、海馬ニュ-ロン、網膜神経節細胞など、数クラスのニュ-ロンの生存と分化をin vitroで促進する事が知られています。
General description
BDNFはNGF、NT-3およびNT-4などの成長因子の神経栄養因子ファミリーのメンバーの1つです。すべての神経栄養因子は6つの保存されたシステイン残基を持ち、アミノ酸レベルで55%の配列の相同性を有しています。BDNFはin vitroで数種のニューロンの生存と分化を促進することが示されています、たとえば神経堤およびプラコード由来の感覚器ニューロン、中脳黒質ドーパミン作動性ニューロン、前脳基底部コリン作用性ニューロン、海馬ニューロン、および網膜神経節細胞などがあります。1 BDNFは抹消神経節内で発現しニューロン標的領域に限定されません。BDNFは神経細胞に対してだけでなく非神経細胞にもパラクリンまたはさらにオートクリン作用も持っている可能性が高まっています。
