本酵素は、成熟ラット脳細胞を使用したコンドロイチン硫酸プロテオグリカンにおける急性の持続的変化の発症の経過を調べる研究に使用されています。コンドロイチナーゼABCは、新鮮なウシ角膜実質から得られた培養角膜実質細胞において、コンドロイチンおよびデルマタン硫酸の定量のための、グリコサミノグリカンの消化に使用されています。

Biochem/physiol Actions

コンドロイチナーゼABCは、4-デオキシ-β-D-グルコ-4-エンウロノシル基を含む二糖に結合した、（1-4）-β-D-ヘキソサミニル、および（1-3）-β-D-グルクロノシルまたは（1-3）-α-L-イズロノシル結合を含む多糖の脱離的分解分解を触媒します。本酵素は、コンドロイチン-4-硫酸、コンドロイチン-6-硫酸、およびデルマタン硫酸に作用し、また、ヒアルロン酸にもゆっくりと作用します。分子用は約120 kDaであり、分子量86 kDaおよび32 kDaの2つの非相同のサブユニットを持っています。最適pHは、コンドロイチン硫酸で8.0、ヒアルロン酸で6.8であり、最適温度は37°Cです。1 mMのZn2+は阻害剤として機能し、一方、0.05 Mの酢酸は酵素を活性化します。

1ユニットは、サメ軟骨由来のコンドロイチン硫酸から、pH 8.0および37°Cで1分あたり1.0 μmolの2-アセトアミド-2-デオキシ-3-O-（β-D-グルコ-4-エン-ピラノシルウロン酸）-4-O-スルホ-D-ガラクトースおよび1.0 μmolの2-アセトアミド-2-デオキシ-3-O-（β-D-グルコ-4-エン-ピラノシルウロン酸）-6-O-スルホ-D-ガラクトースの混合物を遊離させます。
