シグマ社の酵素は、固定化金属イオンアフィニティクロマトグラフィー（IMAC）および固定化金属イオンアフィニティキャピラリー電気泳動（IMACE）を用いたグリコシル化α-キモトリプシンの構造活性相関の研究に用いられています。

Biochem/physiol Actions

α-キモトリプシンはジスルフィド結合により結合した3つのポリペプチド鎖（A鎖-13残基、B鎖-131残基、C鎖-97残基）からなる、計241アミノ酸残基を持つセリンペプチダーゼです。この酵素の分子量は25 kDaであることがわかっています。plは8.75です。C末端側のチロシン、フェニルアラニン、トリプトファンおよびロイシンのペプチド結合を選択的に加水分解します。Ca2+はこの酵素を活性化および安定化します。この酵素は以下の化合物等により阻害されます：ジイソプロピルフルオロリン酸（DFP）、フェニルメタンスルホニルフロリド（PMSF）、N-p-トシル-L-フェニルアラニンクロロメチルケトン（TPCK）、キモスタチン、アプロチニン、α1-アンチトリプシンおよびα2-マクログロブリン、10 mM Cu2+およびHg2+。
結合のカルボキシル基側にに芳香族側鎖または大きな疎水性側鎖(チロシン, トリプトファン, フェニルアラニン, メチオニン)を持つペプチド結合を加水分解するセリンプロテア-ゼです。

1ユニットは、pH 7.8、25°Cで1分間あたり1.0 μmolのBTEEを加水分解します。
