General description

Taqポリメラーゼは、耐熱細菌のThermus aquaticusに由来する耐熱性のDNAポリメラーゼです。安定なデオキシリボ核酸（DNA）ポリメラーゼ（至適温度80°C）は、高度好熱菌であるThermus aquaticusから精製されました。本酵素は、ホスホモノエステラーゼ、ホスホジエステラーゼ、および一重鎖エキソヌクレアーゼ活性を含んでいません。本酵素の最大活性には、4つすべてのデオキシリボヌクレオチドと、活性化されたウシ胸腺DNAが必要です。

Application

Thermus aquaticus由来のTaq DNAポリメラーゼは、以下の用途に使用されています：

Biochem/physiol Actions

Taqポリメラーゼは、オリゴヌクレオチドプライマーによる、DNAテンプレート依存的な、相補的DNA鎖へのdNTPの取り込みを触媒します。本酵素は、5'から3'のポリメラーゼおよびエキソヌクレアーゼ活性の両方を示します。

Taq DNA polymerase は、高温細菌Thermus aquaticusから分離された専用の熱安定性酵素です。大腸菌で発現したリコンビナント酵素です。SDS-PAGEで検出したところ、この94kDaのタンパク質にエンドヌクレア-ゼ・エキソヌクレア-ゼのコンタミネ-ションは認められません。この酵素は、5′→3′ポリメラ-ゼ活性とエキソヌクレア-ゼ活性の両方を有しています。
