Biochem/physiol Actions

ECMゲルは、20～40℃になると熱活性型の重合を受け、再構成基底膜を形成します。@°@ PC12細胞は、このECMゲルの薄層上で成長した場合、2日以内に軸索形成を示します。

本製品の製造に使用するすべてのマウスコロニーは、複数の病原体について日常的にスクリーニングされています。LDEVが陰性であることが試験され明らかにされています。

Preparation Note

この製品は、マウスのEngelbreth-Holm-Swarm肉腫から調製されます；クロロホルムに対して透析。ゲルは、使用前に2～8℃で一晩解凍したのち、2～8℃に予冷したプレートとピペットを使用してマルチウォールプレートに分注する必要があります。ゲルは、2～8℃のDulbecco改変Eagle培地で最大2倍に希釈可能で、ゲルをプレートに加える前に行う必要があります。 本製品は、20℃で5分以内にゲル化します。長時間操作する場合は、10℃未満で作業を行う必要があります。細胞は、0.5 mmの薄いゲル層の表面に播種するか、または1 mm層の中で培養できます。 層内で細胞を培養する場合は、1 mLあたり3～4 x 104細胞の推奨密度で播種する前に、細胞をゲルに添加します。 このゲルから細胞を解離するためには、カルシウム、マグネシウム、またはEDTAを含有していないプロテアーゼのPBS溶液を、1 mLあたり0.6～2.4単位の濃度で使用します。

Other Notes

ECMは、主成分としてのラミニン、IV型コラーゲン、ヘパリン硫酸プロテオグリカン、エンタクチン、その他の微量成分を含有し、タンパク質濃度が8～12 mg/mL（DMEM中）となるように調製されました。ECMゲルにIV型コラーゲンを添加すると、重合が増加します。

製品には、50 mg/L硫酸ゲンタマイシンが含有されています。

Disclaimer

長期間保存する場合は、製品を-20℃に維持してください。ECMゲルは、2～8℃で72時間まで保存できます。
