本酵素は、ヌクレアーゼ、ベンゾナーゼによる分解から保護されているTf1 RNAの量を検出するアッセイに使用されています。Schizosaccharomyces pombeのTf1エレメントは、機能的プロテアーゼ、逆転写酵素、およびインテグラーゼタンパク質をコードする長い末端反復配列を含むレトロトランスポゾンです。これは、細胞溶解物から得られる核ペレットおよび核抽出物の分画にも使用されています。
超高純度ベンゾナーゼヌクレアーゼ、またはSerratia marcescens（セラチア／霊菌）由来のエンドヌクレアーゼは、Serratia marcescensのエンドヌクレアーゼ活性に及ぼすドデシルベンゼンスルホン酸ナトリウムおよび消泡剤の効果を評価する研究に使用されています。Serratia marcescens由来のエンドヌクレアーゼは、クロマチンサブユニットの構造を調べる研究にも使用されています。

Biochem/physiol Actions

ベンゾナーゼ®は、Serratia marcescens（セラチア／霊菌）が由来の遺伝子組換えエンドヌクレアーゼです。このタンパク質は、不可欠な2つのジスルフィド結合を有する30 kDaサブユニットのダイマーです。このエンドヌクレアーゼは、すべての形態（一本鎖、二本鎖、直鎖状および環状）のDNAとRNAを攻撃および分解し、広範囲の操作条件にわたって効果的です。これは、核酸を5'-一リン酸塩末端を有する3～5塩基長のオリゴヌクレオチドに断片化します。これは、組換えタンパク質から核酸を除去するのに理想的です。また、核酸を完全に断片化することが望ましい用途にも使用できます。さらに、タンパク質抽出物の粘度を低下させ、細胞凝集を防止します。この酵素によるタンパク質サンプルの前処理をすれば、結合した核酸をすべて排除することにより、2Dゲル電気泳動での分解能が向上します。酵素活性の最適pHは、8.0～9.2であることが判明しています。
未変性または熱変性のDNA, RNAを分解します。

Other Notes

1ユニットは、反応液を2.625 mLとした場合に、pH 8.0、37°C、30分間で、超音波破砕されたサケ精子DNAを切断して、1.0のΔA260に相当する酸可溶性オリゴヌクレオチドを生じる酵素量です。
