本モノクローナル抗体は N 末端の FLAG シーケンスを認識します。

General description

エピトープタグは、さまざまな種類の細胞内で遺伝子産物の位置を特定するため、タンパク質およびタンパク質複合体のトポロジーを研究するため、関連タンパク質を同定するため、またタンパク質特異的抗体がない場合に新たに同定された少量または低免疫原性のタンパク質の特徴付けを行うために使用できます。標的タンパク質のN末端、メチオニン残基が先行するN末端、C終端、または内部に対してFLAG®ペプチド配列によるタグ付けを行えます。また、FLAGを他のタグと関連させて配置することもできます。FLAGタグまたはFLAG配列はサイズが小さく、親水性が高いため、タンパク質発現、タンパク質分解的成熟、抗原性および機能を妨害する可能性は低い傾向にあります。N末端FLAG®ペプチド配列には独特なエンテロキナーゼ切断部位が含まれるため、精製融合タンパク質から完全に除去できます。融合タンパク質からのCu2+イオン触媒C末端FLAG®ペプチド切断が報告されています。FLAG®ペプチドと同一の、8つのアミノ酸残基のうち5つの残基を含む配列モチーフが、ラットおよびマウスのMg2+依存性タンパク質b-ホスファターゼのほか、ヒトおよびウシの酵素で確認されています。ウサギANTI-FLAG®抗体は、ハイブリドーマSIG1-25から産生されます。このハイブリドーマは、KLHと抱合したFLAG®ペプチド配列で免疫化したウサギから得た骨髄腫細胞と脾細胞を融合することで作製されます。
