General description

ヒストンタンパク質は核DNAを安定化させ、調節することが知られています。コアヒストンであるH2A、H2B、H3、H4は、真核生物の核DNAの反復ヌクレオソーム単位へのパッケージングを助け、リンカーヒストンであるH1はヌクレオソーム間でDNAに結合します。 ヒストン、特にヒストンのN末端尾部ドメイン、特にH3とH4では、アセチル化、リン酸化とメチル化、ユビキチン化、ADPリボシル化など、多様で精巧な一連の翻訳後修飾が生じます。これらの修飾がクロマチン状態と遺伝子発現を決定づけています。
主にSer10とSer28におけるH3のリン酸化は、高度にアセチル化されたH3のごく一部に局在し、主に分裂促進およびストレス刺激に応答して発生します。Ser10のリン酸化は有糸分裂の開始に重要な事象であると考えられています。これは、有糸分裂と減数分裂の両方における染色体の凝縮と密接に相関しています。ヒストンのメチル化は、アセチル化と同様に、複雑で動的なプロセスであり、転写調節、クロマチン縮合、有糸分裂とヘテロクロマチンの構築など、多くのプロセスに関与しています。ヒストンH3、Lys4、Lys9、Lys27のN末端尾部ドメインに保存されているリシン残基は、メチル化の好発部位です。ヒストンリシン残基は、さまざまなヘテロクロマチンサブドメインでモノメチル化、ジメチル化、トリメチル化されている可能性があり、クロマチン構造の調節にさらなる複雑さを加えています。
