General description

ヒアルロニダーゼは、ヒアルロン酸代謝酵素です。これは、ヘビ、ミツバチおよび膜翅目昆虫の毒液中に認められます。ヒアルロニダーゼはヒアルロン酸（HA）を分解し、それによって膜の透過性を高め、粘性を低くし、組織透過性を高めます。これは、局所麻酔薬の拡散を促進し、心筋梗塞のサイズを減少させ、抗がん剤の効果を向上させ、自然分娩時の会陰部外傷を防ぎます。

Application

ヒツジ精巣由来のヒアルロニダーゼは、下記の用途で使用されています：
  • フローサイトメトリー
  • 推定上の精原幹細胞（SSC）の単離
  • 磁性ヒドロゲル特性における磁気刺激の影響の研究
  • II型コラーゲンの分布を可視化する免疫組織化学での使用
  • パッチクランプ法のためのミューラー器官のIII群神経細胞の露出
  • ヒドロゲルの変性

これらの酵素は、ヒアルロン酸、コンドロイチン、および硫酸コンドロイチン内のβ-N-アセチルヘキソサミン-[1→4] グリコシド結合をランダムに切断します。

Enzymatic Assay of Hyaluronidase (3.2.1.35)

To measure hyaluronidase activity, a turbidimetric determination assay is used at 600 nm. One unit of hyaluronidase activity will cause a change in absorbance of 0.330 per minute at pH 5.35 at 37 °C.
