Application

キーホールリンペットヘモシアニン（KLH）は実験動物およびヒトに対する強力な、よく知られた免疫刺激物質ですKLHは、抗原特異的T細胞プライミングを強化するためにヘプタン担体タンパク質として使用されており、CD4+T細胞応答を促進することが知られています。ヘモシアニンはTh1応答を促進し、既知の副作用がないことから、癌の長期的な連続的治療に理想的です。

General description

ヘモシアニンは高分子量銅含有糖タンパク質です。ヘモシアニンは可逆的に酸素と結合し、軟体動物で細胞外呼吸タンパク質を生成します。キーホールリンペットヘモシアニン（KLH）は、Megathura crenulata（スカシガイ）の血液リンパをろ過して得られ、巨大キーホールリンペットとも呼ばれます。スカシガイは南カリフォルニアの海岸およびメキシコを原産地とします。
腹足類ヘモシアニンは、自己集合して直径35 nmの中空の円筒を形成する10個のサブユニットを複雑に配置して設計された大きい糖タンパク質（4～8 MDa）です。

Preparation Note

0.46 MのNaCl、2％のPVPおよび41 mMのショ糖を31 mMのリン酸ナトリウムバッファー（pH 7.4）に溶解して乳白色溶液を得るために、KLHを脱イオン化水で溶解して濃度10 mg/mLとします。
タンパク質を溶解した溶液は、-20℃で最大2ヵ月間保存できます。
