Application
SIGMA FASTp-ニトロフェニルリン酸タブレットは、以下の用途に使用されています：
- 非分泌型アルカリホスファターゼを定量して骨芽細胞様細胞（SaoS-2）の分化をモニタリングするため
- アルカリホスファターゼアッセイにおいてマウスの前骨芽細胞であるMC3T3-E1細胞を染色するため
- 酵素結合免疫吸着測定法（ELISA）を用いてヘモグロビン結合α-シヌクレインを測定するExtrAvidin®アルカリホスファターゼの基質として
- アビリン毒素のELISA検出のためのアルカリホスファターゼ結合抗体の基質として
酵素免疫測定法（EIA）におけるアルカリホスファターゼ活性検出用の可溶性基質として開発されました。 pNPP は、高感度のアルカリホスファターゼを用いた EIA アッセイにおける優れた基質です。pNPP タブレット 1 個と Tris バッファータブレット 1 個を脱イオン水に溶解すると、基質溶液としてそのまま使用できます。 各 SIGMAFAST タブレット 1 セットから、pNPP 1.0 mg/mL の pNPP を含有する 0.2 M Tris バッファー溶液が得られます。pNPP を使用した EIA は、経時アッセイとして測定することも、3N NaOH で反応停止させて後から測定することもできます。SIGMAFAST™ は蒸留水または脱イオン水を加えるだけで活性型の基質溶液を調製できる製品です。タブレットはホイルで個別包装されています。
Preparation Note
1錠を5 mLの脱イオン水に溶解すると、そのまま使用可能なpNPP含有バッファ-溶液が得られます。
