Application
ホスファターゼ阻害剤カクテルは、細胞抽出物中に存在するタンパク質修飾酵素の阻害剤を添加することで、インタクトのタンパク質の収量を改善します。 本製品は、ヒト臍帯およびウシ肝臓を含む動物組織由来抽出物、ならびにA431、CHO、およびU937 の細胞抽出物で評価されています。pNPPとアルカリホスファターゼの L-アイソザイムとの比較評価、およびセリン-スレオニンタンパク質ホスファターゼの評価に最適化されています。
Biochem/physiol Actions
本混合物は、特異的な阻害特性を持った個々の化合物を含んでいます。 カンタリジンは、タンパク質ホスファターゼ 2A を阻害します。 （−）-P-ブロモレバミゾールシュウ酸塩は、アルカリホスファターゼの L-アイソフォームを阻害します。 カリクリン A は、タンパク質ホスファターゼ 1 および 2A を阻害します。
Preparation Note
本カクテルは、ジメチルスルホキシドの透明溶液として提供されます。組織または細胞由来タンパク質 500 mg あたり 1 mL のカクテルを希釈することをお勧めします。 最終濃度として 1％（v/v）が使用され、この濃度は、ヒト臍帯およびウシ肝臓由来の 27,000 x g 上清、および A431、CHO、および U937 細胞抽出液の 100,000 x g 上清におけるホスファターゼ活性を阻害します。
