核酸からのタンパク質の除去は、フェノール：クロロホルム：イソアミルアルコールの溶液を用いた抽出により達成できます。核酸の粗混合物については、フェノール：クロロホルム抽出の前に、広範囲のタンパク質分解酵素で断片化することが有益な場合もあります。タンパク質はフェノールにより効率的に変性されるにも関わらず、RNase活性は完全には阻害されません。このため、少量のイソアミルアルコールを添加することにより、RNase活性の不活性化をさらに確実にします。核酸からタンパク質を除去するためのプロトコルについては、製品情報シートをご覧ください。

DNA抽出を行う場合、フェノール相のpHは、トリス緩衝液で平衡化することにより8.0に調整できます。Sigmaは、DNA抽出に特化した製品P2069を提供しています。本製品には、pH調整に便利な平衡緩衝液が含まれています。
