Biochem/physiol Actions

作用機序：ポリミキシンB硫酸塩は、細菌リポポリサッカライド†の脂質A部分と結合し、細菌壁内にヌクレオチドの漏出を可能にするのに十分大きな細孔を誘導することにより細胞膜を破壊します。 これは細胞膜の透過性を破壊します。

抗菌スペクトル：大腸菌（E.coli）を含むほとんどのグラム陰性桿菌††、ならびにほとんどの菌類およびグラム陽性菌に対し殺菌作用を有します。
硫酸ポリミキシンＢは、緑膿菌の感受性株による尿路、髄膜および血流感染症の治療のために臨床的に使用されています。本製品は、多剤耐性病原体を試験する際に、エンドトキシンを除去するための固定化された薬剤として、および切除されたソルガムの根から得られた皮質細胞の膜内で細孔形成を誘導するためにも使用されています。

供給された製品は、2～8℃の暗所で保存する必要があります。 このような条件で3年間保存したサンプルに変化は認められませんでした。 保存溶液は滅菌ろ過し、2～8℃で保存する必要があります。37℃で5日間安定です。

General description

ポリミキシンBは、抗生物質のポリミキシングループに属し、Bacillus polymyxaおよび関連する種のさまざまな株から得られます。硫酸ポリミキシンBには環状ヘプタペプチド単位およびトリペプチド側鎖が含まれており、脂肪アシル残基によりN-アシル化されます硫酸ポリミキシンBは、一般にグラム陰性細菌感染症の治療に使用されます。

Preparation Note

硫酸ポリミキシンＢは50 mg/mLの濃度で水に溶け、ごくわずかに濁りを帯びた無色から黄色の溶液を生成します。 有機溶媒には最小限しか溶けません。 たとえば、エタノールへの溶解度は0.115 mg/mLです。
