百日咳毒素は、ヘテロ三量体グアニンヌクレオチド調節タンパク質Gi、Go、GtのαサブユニットのADPリボシル化を触媒します。 この反応により、Gタンパク質のヘテロ三量体と受容体との相互作用が阻害され、結果としてそれらのカップリングと活性化が阻害されます。 Gαサブユニットが不活性状態でGDP結合に残るため、アデニリルシクラ-ゼや開口K+チャネルは不活性化されません。
