Application

PMSFは、細胞や組織の溶解後にタンパク質を分解するおそれのあるプロテアーゼを阻害することにより、タンパク質の単離中やサンプルの調製中に目的のタンパク質の保存を補助するための溶解バッファーによく使用されます。 PMSFは水性培地中では不安定です。 そのため、水性培地に使用する前に、PMSFの原液を無水有機溶媒（例えば、100%エタノールまたは無水イソプロパノール）で調製するのが一般的です。

PMSFには以下のような一般的特徴や利点があります：
  • トリプシンやキモトリプシンなどのセリンプロテアーゼを阻害します
  • また、システインプロテアーゼ（還元チオールによって可逆）や、哺乳類アセチルコリンエステラーゼも阻害します
  • DFPほどの効果もしくは毒性はありません
  • 有効濃度は0.1～1 mMです
  • 半減期はpH 7.5で1時間
フッ化フェニルメチルスルホニルは、以下のアプリケーションに使用されています：
  • 細胞分画。
  • 核タンパク質抽出における添加剤。
  • コレステロールエステラーゼ（CE）および偽性コリンエステラーゼ（PCE）の阻害剤。
  • 血漿 ANP レベルの定量のための遠心分離前の血液採取。

Biochem/physiol Actions

PMSFを投与すると、その抗コリンエステラーゼ作用と関連しない鎮痛が得られ、中枢に投与したβ-エンドルフィンの鎮痛作用が延長します。

General description

フッ化フェニルメチルスルホニル（PMSF）は、幅広く使用されているセリンプロテアーゼ阻害剤で、キモトリプシン、トロンビンおよびトリプシンなどの酵素に対して有効です。 PMSFは、セリンプロテアーゼの反応部位においてセリン残基のヒドロキシル残基をスルホン化することにより、阻害剤として作用します。
