ポリ-L-リジン・ポリマーは、固体基質への細胞の付着の促進、薬物輸送を増進するためのメトトレキサートへの結合、膵島のマイクロカプセル化、細胞マイクロカプセル化技術、マイクロアレイ・ガラス・スライド・コーティング、および染色体の調製に使用することができます。 低分子量のポリ-L-リジン(30,000～70,000)の溶液は粘性が小さいですが、高分子量のものでは一分子当たりの付着部位が多くなります。

Biochem/physiol Actions

ポリ-L-リジンは、細胞膜の負帯電イオンと培地表面の間の静電気的相互作用を改良して固形基質への細胞接着を促進するのに役立つ、細胞の非特異的付着因子です。 細胞培地表面に吸収されたポリ-L-リジンは、細胞結合に利用できる正帯電部位の数を増やすように機能します。 ポリ-L-リジンを消化できる細胞と共に、ポリ-D-リジンは付着因子として使用されます。

Preparation Note

本製品の分子量は15,000～30,000です。HBrを除去するためには、本製品を中性緩衝液中で溶解し、透析して塩を除去します。 ポリ-L-リジン製品はトリフルオロ酢酸にさらしておらず、透析してすべての単量体、二量体、三量体を除去します。薄層クロマトグラフィーによって確認済みです。 概して、本製品を付着因子として使用するためには、50 mLの無菌組織培養グレード水を5 mgのポリリジンに加え、25 cm2当たり1 mLの溶液で表面を無菌状態でコートします。 5分後に、吸引によって溶液を除去し、表面をよく洗浄します。 細胞と培地を導入するまで2時間乾燥させます。

Other Notes

ポリ－L-リジンはリジン残基当たりおよそ1個のHBrを有する正に荷電したアミノ酸ポリマーです。臭化水素酸塩により、ポリ-L-リジンを水溶性の結晶形態にできます。HBrがアミノ基と、カルボキシル基またはNあるいはO含有部分のいずれかとの間の水素結合を妨害するため、製品中に少量のβ構造が含まれることがあります。

滅菌溶液は、2 ～ 8°Cで保存した場合、最大 2 年間安定です。-20°Cでは乾燥させて保存してください。
