Biochem/physiol Actions
Laemmli サンプルバッファーは、Laemmli の SDS-PAGE システムにおいて使用されるタンパク質サンプル用に特別に作成されています。 タンパク質サンプルを2倍サンプルバッファー（1 ： 1）と混合して、沸騰した水で2 ～ 5 分間加熱します。2-メルカプトエタノールは、分子内および分子間のジスルフィド結合を還元します。 SDS はタンパク質やサブユニットを変性し、それらの全体にマイナス電荷を付与することで、サイズに基づく分離を可能にします。 ブロモフェノールブルーは、タンパク質の泳動先端を示す色素として使用され、また、ローディングの際にサンプルを可視化する役割も持っています。 グリセロールは、サンプルの密度を上げることで、サンプルウェルにおいてサンプル層を形成します。
Other Notes
溶液は、4%SDS, 20%グリセロ-ル, 10% 2-メルカプトエタノ-ル, 0.004%ブロモフェノ-ルブル-, 0.125M Tris-HCl(pH 約6.8)を含有しています。
Preparation Note
使用例:サンプルに等容量のゲルロ-ディング溶液を添加し、十分に混合してください。
