Biochem/physiol Actions
トリプシンは、リジンおよびアルギニン残基のC末端側のペプチドを切断します。酸性残基が切断部位の両側にある場合、この反応の加水分解速度は遅くなり、プロリン残基が切断部位のカルボキシル側にある場合、加水分解は停止します。トリプシン活性の至適pHは7～9です。また、トリプシンは、アミノ酸の合成誘導体のエステルおよびアミド結合を切断するよう作用します。EDTAは、トリプシンが作用するペプチド結合を不明瞭にするカルシウムおよびマグネシウムイオンを中和するキレート剤としてトリプシン溶液に添加されます。これらのイオンを除去すると、酵素活性が増加します。

Disclaimer
1 mM HCl中の溶液は、アリコートで1年間安定であり、-20°Cで保存します。また、Ca2+が存在すると、トリプシンの自己分解が低減されて溶液中での安定性が維持されます。トリプシンは2.0 M尿素、2.0 MグアニジンHCl、または0.1%（w/v）SDS中でもその活性の大部分を保持します。

トリプシンは、223アミノ酸残基の単鎖ポリペプチドから構成されます。トリプシノーゲンのLys-lleペプチド結合が切断されてN末端のヘキサペプチドが取り除かれるとトリプシンになります。そのアミノ酸の配列は、6つのジスルフィド結合によって架橋されます。これは、天然型のトリプシンであるベータトリプシンです。ベータトリプシンは、自己分解によりLys-Ser残基で切断されるとアルファトリプシンになります。トリプシンはセリンプロテアーゼファミリーのメンバーです。
基質として1単位のBAEEを使用したとき、pH 7.6、25°CにおけるA253は0.001/分となります。
