General description

Sigma 7-9® は、トリズマ塩基またはトリス（ヒドロキシメチル）アミノメタンとしても知られる汎用性の高い生理的緩衝液であり、タンパク質凍結、電気泳動、および TAE や TBE などの緩衝液の調製といったさまざまな用途に使用されています。トリスはpKaが8.06であるため、生体の典型的な生理的pHの範囲（pH 7.0～9.0）で行う実験には特に有用です。溶液のpHを微調整するために、他の緩衝剤と組み合わせて使用することもよくあります。

さらに、トリスは細胞膜の浸透性を向上させるためにも役立ちます。この化合物は、生体液と適合性のあるバッファーを調製するためにこれを使用している研究室にとっては不可欠であり、標準pH溶液としての役割を果たし、また乳酸脱水素酵素アッセイ、in situハイブリダイゼーション、および細胞からのタンパク質抽出などの手順を容易にします。

Other Notes

すべてのバッファーの pH は、温度や濃度に依存します。トリスバッファーのpHは、温度が 1°C下がるごとに0.03 上昇し、10 倍希釈によって約 0.03 ～ 0.05 減少します。
厳密なアプリケーションでは、適切に校正した、ガラス/甘汞電極の pH メーターを使用してください。
