Application

N-p-トシル-L-フェニルアラニンクロロメチルケトンは、以下に使用されています。

特定のセリンプロテアーゼの不可逆的インヒビター。トリプシン標品中のキモトリプシンの阻害に用いられます。

Biochem/physiol Actions

LPSまたはサイトカインが誘導する核因子κB(NF-κB)の活性化を遮断し、その結果、iNOSおよびCOX-2の転写誘導も遮断します。すべてのマイトジェンによるpp70s6kの活性化をブロックします。一部の細胞株で刺激に応答して起こるカスパーゼのプロセシングを阻害することによりアポトーシスを遮断します。タキソールによって開始されたアポトーシスも阻害します(ヒト乳癌細胞MCF-7)。
N-p-トシル-L-フェニルアラニンクロロメチルケトン（TPCK）は、リポポリサッカライド（LPS）またはサイトカインによる核因子κB (NFκB)の活性化をブロックし、それによって誘導性一酸化窒素合成酵素（iNOS）およびシクロオキシゲナーゼ-2（COX-2）転写の誘導がブロックされます。すべてのマイトジェンによるpp70s6kの活性化をブロックします。一部の細胞株および一部の刺激に対するカスパーゼの処理を阻害することにより、アポトーシス細胞株をブロックします。タキソール（MCF-7ヒト乳がん細胞内）によって開始されるアポトーシスもブロックします。 TPCKは、キモトリプシンのフェニルアラニン残基を認識し、その機能を妨げます。
