Application
この製品は、細胞培養用途について検査されています。 トリプシンは、Vibri sp. V26の非毒素産生マングローブ分離株由来のアルカリプロテアーゼについて動物細胞培養における潜在的用途を評価する試験に使用されています。 トリプシンは、好酸球ムチン中の真菌の検出を改善する研究にも使用されています。
Biochem/physiol Actions
このインヒビターはトリプシンに対して作用し、キモトリプシンおよびプラスミンに対してこれよりも弱い作用を及ぼします。 このインヒビターはトリプシン、血漿カリクレイン、凝固因子Ｘと同様のメカニズムをもってプロテアーゼも阻害します。トリプシンインヒビターは、メタロプロテアーゼ、組織ベースのカリクレイン、酸プロテアーゼまたはチオプロテアーゼに対しては作用しません。 このインヒビターは、プロテアーゼ活性部位の1：1の化学量論合成物を生成することで作用し、次にインヒビターの単一のアルギニン-イソロイシン結合が開裂します。 阻害は可逆性でpH依存性です。
Preparation Note
トリプシンインヒビターは水およびリン酸バッファーに10 mg/mLの濃度で溶けます。 また、平衡塩溶液に1 mg/mLの濃度で溶け、無血清培地にも溶けます。 濃度が10 mg/mLを超える濃縮溶液は濁ることがあり、黄色からアンバーを呈します。 細胞をトリプシン化した後、解離に使用したトリプシン溶液1 mLに対し1 mLのトリプシンインヒビター溶液に1 mg/mLで再懸濁させます。 次にこの細胞懸濁液を1000 rmpで遠心分離し、細胞ペレットを生成します。
溶液は2～8℃で短期間保存したとき活性を保持できます。溶液はアリコートを-20℃で凍結させると安定します。

Analysis Note
1 mgのトリプシンインヒビターで、タンパク質1 mg当たりの活性が約10,000 BAEE単位のトリプシン1.0～3.0 mgが阻害されます。
Other Notes
BAEEを基質として使用すると、トリプシン1単位で、pH 7.6、25℃、反応容量3.2 mL、光路1 cmにおいてδＡ253が0.001/分となります。
ダイズトリプシンインヒビターは、2つのジスルフィド結合で架橋されたアミノ酸残基181個を有する単一のポリペプチド鎖を含む一量体タンパク質です。
