Application
鶏の卵白由来のトリプシン阻害剤は以下の用途に用いられています：
- 変性され、酸化され、脱グリコシル化されたオボムコイドを産生するため、および患者由来の細胞培養をT細胞エピトープのマッピングのために刺激するため
- フローサイトメトリー分析のために核を処理するため
- プラナリアの細胞への解離に用いるホルトフレーター溶液
General description
オボムコイドの25%は糖質が占めています。
オボムコイドまたはトリプシンインヒビターは、大部分の鳥類の卵白に豊富に存在するタンパク質です。鶏卵タンパク質は、約186個のアミノ酸残基からなり、高度にグルコシル化されています。3つのタンデムドメインはそれぞれ、膵臓トリプシンインヒビターと相同です。極めて免疫原性が高く、おそらく卵アレルギーの大部分の症例の原因であると考えられます。
Other Notes
1トリプシンユニットは、BAEEを基質として用いる場合、pH 7.6、25°C、1分間に、ΔA253が0.001となる酵素量です(反応液量 = 3.2 mL、光路長 = 1 cm)。
ニワトリ卵白由来のオボムコイドは、それ自体はキモトリプシンを阻害しません。キモトリプシンの阻害は、Feeney, R. E., et al., J. Biol. Chem., 238, 1415 (1963)の方法により、オボインヒビタ-混入の目安となります。
