General description

Ubiquitin carboxyl-terminal hydrolase isozyme L1 (UniProt: P09936; also known as UCH-L1, Neuron cytoplasmic protein 9.5, PGP 9.5 (PGP9.5), Ubiquitin thioesterase L1) is encoded by the UCHL1 gene (Gene ID: 7345) in human. Protein gene product 9.5 (PGP9.5) is the historical name for ubiquitin C-terminal hydrolase L1 (UCHL1), a cysteine protease of the UCH deubiquitinase family that is highly enriched in neurons and widely used as a pan-neuronal/neuroendocrine marker in histopathology. Structurally, PGP9.5 adopts the canonical UCH fold and contains a catalytic Cys–His–Asp triad (human Cys90/His161/Asp176), with crystallographic analyses showing conformational features of the active site and ligand-induced rearrangements upon binding ubiquitin-based inhibitors/substrates that help rationalize its substrate selectivity and catalytic tuning. PGP9.5 participates in ubiquitin homeostasis by hydrolyzing C-terminal adducts from ubiquitin and processing ubiquitinated proteins, thereby interfacing with the broader ubiquitin–proteasome system (UPS) that governs proteostasis in long-lived cells such as neurons. PGP9.5 protein is strongly detected across neuronal cell types, predominantly localized to the cytosol with a substantial fraction reported as membrane-associated in brain, consistent with roles at neurites/synapse-proximal compartments and dynamic protein quality control. PGP9.5 has been discussed in the context of Parkinson’s disease genetics (e.g., the rare familial I93M variant and the debated S18Y polymorphism) and in neurodegeneration models implicating altered PGP9.5 enzymatic activity and downstream metabolic/proteostatic consequences.

Preparation Note
