Western blot: Identifies ~ 200kDa band in reduced samples. Use Tris based buffers for best results as epitope is phosphate dependent; Different regions of the CNS were dissected out immediately and homogenized in BUST buffer (8 M urea, 0.5% SDS, 2% 13-mercaptoethanol, and 50 mM Tris, pH 7.4) at a ratio of i ml/100 mg wet tissue. The tissue suspensions were sonicated well and centrifuged in a TL-100 ultracentrifuge (Beckman Instruments, Fullerton, CA) at 100,000 g
for 1 h at 25°C, and the protein concentrations in the supernatants were determined. Proteins were separated on 7.5% SDS-PAGE gels, {Tu. P et al (1995) J Cell Biol 129:1629-1640}
Immunohistochemistry on frozen sections; not effective on paraffin tissues in original work. {Carden et al, 1987}.
Optimal working dilutions must be determined by end user.

Biochem/physiol Actions
Neurofilament H (NF-H), carboxy terminal (phosphate dependent (+). Clone is specific for phosphorylated NF-H {Carden et al (1987) 7(11):3489}.
Physical form
Unpurified tissue culture supernatant from a perfusion system, filtered through a 0.2μ membrane prior to vialing. Product contains 20%FBS and Ciprofloxacin at final concentration of 10μg/mL.
Preparation Note
Maintain at -20°C in undiluted aliquots for up to 12 months after date of receipt. Avoid repeated freeze/thaw cycles.
